Ionophores enable the transport of Na+ ions in cells and promote the intracellular concentration of Na+ ions. This increased concentration of Na+ ions inhibits certain mitochondrial functions, such as substrate oxidation and ATP hydrolysis. Intracellular Na+ ion exchanges for extracellular Ca++ and improved intracellular concentration of calcium ions lead to cytotoxicity. In addition, some drugs directly facilitate Ca++ transport in cells, and increased intracellular concentrations of Ca++ in cardiac and skeletal muscle cells are responsible for its toxic effects in cells.
